Effects of Mild Moxibustion on TNF-α and IL-10 in Rats with IBS-D Induced by Liver-stagnation and Spleen-deficiency
Objective To observe the therapeutic effect of mild moxibustion on rats with irritable bowel syn-drome with predominant diarrhea(IBS-D)induced by liver-stagation and spleen-deficiency,and to detect the content of tumor necrosis factor-α(TNF-α)and interleukin-10(IL-10)in rats.Methods A total of 24 wistar male rats were randomly divided into a normal group,a model group,a western medicine group and a mild mox-ibustion group,with 6 rats in each group.Except the normal group,the other groups were treated with ice acetic acid enema+restraint stress to establish the IBS-D rat model with liver-depression and spleen-deficiency.Af-ter modeling,the western medicine group was given Piverium bromide intragastric administration(15 mg/kg),once a day.In the mild moxibustion group,acupuncture points of Tianshu,Shangjuxu and Taichong were select-ed for mild moxibustion treatment.The normal group and model group only grasp and bind,and do not do other intervention.After the intervention,the general condition of each group was observed.The rate of thin stool(LSR),the minimum volume threshold of the abdominal retract reflex(AWR)and the open field test were de-tected;the changes of colon histopathology were observed;and the conent of TNF-α and IL-10 in serum and colon tissues were detected.Results(1)Compared with the normal group,the body mass and the minimum volume threshold of AWR in the model group were both decreased while the LSR was increased(P<0.05).Compared with the model group,the body mass and the minimum volume threshold of AWR in the western med-icine group and mild moxibustion group were increased,and LSR was decreased(P<0.05).(2)Compared with the normal group,the total number of perforation,upright times and modification times in the model group were reduced(P<0.05);Compared with the model group,the times of western medicine group and mild moxi-bustion group were increased(P<0.05).(3)Compared with the normal group,the content of TNF-α in serum and colon tissue of model group was significantly increased(P<0.01),and the content of IL-10 was signifi-cantly decreased(P<0.01);Compared with the model group,the content of TNF-α in serum and colon tissue of western medicine group and mild moxibustion group was significantly decreased,and the content of IL-10 was increased(P<0.05).Conclusion Mild moxibustion can relieve the abdominal pain and diarrhea,improve the mental state of IBS-D model rats with liver-stagnation and spleen-deficiency.The mechanism may be relat-ed to reducing TNF-α and increasing IL-10 level,correcting the imbalance of inflammatory factors,and thus enhancing immune function.
