红大戟种子检验规程研究

Study on Inspection Procedures of Knoxia roxburghii Seeds

为了研究红大戟种子检验规程,为红大戟种子质量标准的制定提供依据,参照农作物种子检验规程的方法,通过对红大戟种子扦样、净度分析、种子真实性、千粒重、含水量、发芽率、生活力等常规指标进行对比分析,找出适合红大戟种子的检验方法.结果表明,红大戟种子净度分析试样不少于 0.3 g;采用外观形态观察鉴定法鉴定种子真实性,千粒法测定种子重量;131℃下烘 6h测定种子含水量,采用浓度为 0.1%的TTC溶液 25℃或TTC浓度 0.5%,温度 20~25℃条件下染色 16h测定种子生活力;种子消毒后 25℃下(12h光照/12h黑暗)培养测定种子发芽率,发芽床为沙上.
To study the seed inspection procedures of Knoxia roxburghii,and provide the basis for the formulation of seed quality standards of K.roxburghii.With reference to the method of crop seed in-spection procedures,the conventional indicators such as seed sampling,purity analysis,seed authen-ticity,1 000-grain weight,water content,germination rate,and viability were compared and analyzed to find a suitable test method for K.roxburghii seeds.The results showed that the purity analysis sample of K.roxburghii seeds was not less than 0.3 g,the seed authenticity was identified by mor-phological observation and identification method,and the seed weight was determined by thousand seed weight method.The seed moisture content was determined by baking at 131℃for 6 h,and the seed viability was determined by staining with TTC solution at a concentration of 0.1%at 25℃or TTC concentration of 0.5%at 20-25℃for 16 h.The seed germination rate was measured at 25°C(12h light/12h dark)after seed disinfection,and the germination bed was sand.
