Effects of Hydrogen Peroxide and Ascorbic Acid on the Growth of Mycelia and Sclerotia of Polyporus umbellatus
In order to provide theoretical basis for the standardization of Polyporus umbellatus,the factory production and the formation mechanism of sclerotia.By preparing different concentrations of hydrogen peroxide and ascorbic acid solution,adding to plate medium and bag material culture respec-tively,observed and compared the growth of mycelia and sclerotia.In plate culture,moderate concen-tration of ascorbic acid(1.0 mg/mL,2.0 mg/mL,4.0 mg/mL,5.0 mg/mL)and hydrogen peroxide(0.10%,0.20%,0.40%,0.50%,0.75%,1.0%)could promote the formation of sclerotia.With the increase of concentration,the more dense the mycelium entanglement,the concentration was inverse-ly proportional to the growth rate of mycelium.In the bag material culture,ascorbic acid inhibited the growth of mycelia,and hydrogen peroxide promoted the growth of mycelia.Under certain concentra-tion(hydrogen peroxide:0.40%;ascorbic acid:4.0 mg/mL)could promote the formation of sclero-tia.In conclusion,when the oxidative stress of mycelia growing in plate culture reach a certain level could promote the differentiation of mycelia into sclerotia.In the process of bag material culture,when the ascorbic acid and hydrogen peroxideat at appropriate concentration can promote sclerotia dif-ferentiation.
