Extraction and Antioxidant Activity Analysis of Total Flavonoids of Polygonatum sibiricum
Ultrasonic method was used to extract the total flavonoids of P.sibiricum.Single factor experiments were con-ducted with ethanol volume fraction(A),material liquid ratio(B),ultrasonic time(C),and ultrasonic temperature(D).On the basis of it,Box Behnken Design of experiments was used to optimize the extraction process with the total fla-vonoids extraction rate as the response value;Analyze the antioxidant capacity of each component using DPPH,ABTS,and reducing power measurements.The results showed that the optimal extraction process parameters for total flavonoids in P.sibiricum were as follows:the solid-liquid ratio was 1:48.42 mL·g-1,the extraction time was 50.00 minutes,the ethanol concentration was 53.33%,and the temperature was 54.66 ℃.Under these conditions,the extraction rate of total flavonoids was 30.12+0.03 mg·g-1,which was close to the predicted value,indicating that the response surface model had good predictive ability;The scavenging ability of total flavonoids from P.sibiricum to DPPH and ABTS free radicals shows an increasing trend within a certain range,with a dose-dependent effect.At mass concentrations of 2 mg·mL-1 and 3 mg·mL-1,the scavenging ability of ABTS free radicals is equivalent to that of vitamin C.The total flavonoids from P.sibiricum also have good reducing power,indicating good antioxidant activity and can be used as a good natural antioxi-dant.
