川陈皮素对舒芬太尼麻醉后大鼠认知及学习记忆功能损伤的影响机制

目的 探索川陈皮素对舒芬太尼麻醉后大鼠认知功能及学习记忆功能损伤的影响及作用机制.方法 选取SPF级SD大鼠 65 只,对照组 15 只,50 只建立舒芬太尼模型,其中 45 只成功建模,将建模后大鼠随机分为模型组、川陈皮素低剂量组、川陈皮素高剂量组,每组各 15 只.对照组、模型组给予 5mL生理盐水灌服,川陈皮素低剂量组给予生理盐水+25mg/kg川陈皮素灌服,川陈皮素高剂量组给予生理盐水+55mg/kg川陈皮素灌服.观察各组大鼠逃生潜伏期、空间探索能力,测定超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、Toll样受体-4(TLR4)、核因子-κB(NF-κB)的表达水平.结果 与对照组相比,模型组、川陈皮素低剂量组、川陈皮素高剂量组平台所在象限停留时间、穿越平台次数、SOD、CAT水平表达降低,而逃生潜伏期、MDA、TNF-α、IL-1β、TLR4、NF-κB p65 水平表达升高(P<0.05);与模型组相比,川陈皮素低剂量组、川陈皮素高剂量组逃生潜伏期、MDA、TNF-α、IL-1β、TLR4、NF-κB p65 表达水平降低,平台所在象限停留时间、穿越平台次数、SOD、CAT表达水平升高(P<0.05);与川陈皮素低剂量组相比,川陈皮素高剂量组逃生潜伏期、MDA、TNF-α、IL-1β、TLR4、NF-κB p65 表达水平降低,平台所在象限停留时间、穿越平台次数、SOD、CAT表达水平升高(P<0.05).结论 川陈皮素可改善舒芬太尼麻醉后大鼠认知功能及学习记忆功能的损伤,其机制可能与调节相关因子表达水平有关.
Mechanisms of the Effect of Chpicin on the Impairment of Cognitive and Learning Memory Functions in Rats after Sufentanil Anesthesia
Objective To analyze the effects of Chpicin on cognitive function and learning and memory function in rats after Sufentanil anesthesia.Methods A total of 65 SPF SPRague-dawley rats were selected,15 of which were used as the control group,and the remaining 50 rats were established Sufentanil model.Finally,45 rats were successfully modeled.The 45 rats successfully modeled were randomly divided into model group,low dose group and high dose group,with 15 rats in each group.After successful modeling,The control and model groups were given 5 mL of saline for gavage,the low dose group was given saline+25mg/kg of Chpicin for gavage,and the high dose group was given saline+55 mg/kg of Chpicin for gavage.The escape latency,spatial exploration ability,superoxide dismutase(SOD),catalase(CAT),malondialdehyde(MDA),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),Toll-like receptor-4(TLR4),and the expression levels of nuclear factor-κB(NF-κB)were observed.Results Compared with the control group,the dwell time and crossing times of platform in model group,low dose and high dose group decreased,while the expression levels of SOD,CAT and IL-1β,MDA,TNF-α,TLR4 and NF-κB P65 were increased(P<0.05).Compared with model group,the escape latency,the expression levels of MDA,TNF-α,IL-1β,TLR4 and NF-κB P65 were decreased in low dose group and high dose group,while the dwell time of platform quadrant,the number of crossing platform,SOD and CAT were increased(P<0.05).Compared with the low dose group,the es-cape latency,MDA,TNF-α,IL-1β,TLR4 and NF-κB P65 expression levels in the high dose group were decreased,and the dwell time of platform quadrant,the number of crossing plat-form,SOD and CAT expression levels were increased(P<0.05).Conclusion The use of Chorin improved the impairment of cognitive function and learning and memory function in rats after Sufentanil anesthesia.
