苍术、麸炒苍术有效成分含量测定的研究

目的 就苍术、麸炒苍术中的主要化学成分苍术素、苍术酮、桉叶醇、茅术醇 4 种有效成分含量建立同时测定的方法,以提供苍术、麸炒苍术的质量评价依据.方法 将流动相、固定相、柱温、流速等因素综合考虑,经过多次优化,最后确定最佳色谱条件为:柱温 25℃;检测波长 320 nm;电子分析天平分别称取适量苍术素、苍术酮、桉叶醇、茅术醇、丹皮酚对照品,加甲醇制成溶液,浓度分别为丹皮酚 12 μg/mL,茅术醇 18 μg/mL,桉叶醇 5.5 μg/mL,苍术酮 17 μg/mL,苍术素 450 μg/mL.以冷浸和超声两种方法分别处理 30 min,比较峰面积.采用超声提取法,比较不同时间段甲醇溶剂提取液峰面积.结果 供试品中苍术酮、苍术素、桉叶醇、茅术醇峰面积在各个时间点稳定性较好.苍术炮制后挥发油含量明显下降,而茅术醇和桉叶醇含量下降也很明显.结论 苍术炮制后挥发油中的苍术素、茅术醇和桉叶醇等的升高趋势可能与苍术麸炒后健脾作用增强而燥性减弱有关.
Determination of Effective Components in Rhizoma Atractylodis Chinensis and Rhizoma Atractylodis with bran-stired
Objective To establish a method for simultaneous determination of four main chemi-cal components in Rhizoma atractylodis chinensis,atractylosin,Cineole,and catechol in Rhizoma atractylodis and Rhizoma atractylodis bran-stired,in order to provide the quality evaluation basis of Rhizoma atractylodis and Rhizoma atractylodis bran-stired.Methods The mobile phase,sta-tionary phase,column temperature and flow rate were considered together.After several optimi-zations,the optimal chromatographic conditions were as follows:column temperature,25℃;de-tection wavelength,320 nm;and the control products of atractylodin,atractylodinone,eucalyptol,maitake alcohol and danpihenol were weighed on an electronic balance and made into a solution by adding methanol with the concentrations of danpihenol(12 μg/mL),atractylodinone(17 μg/mL),atractylodinone(450 μg/mL),atractylodinone(17 μg/mL)and atractylodinone(450 μg/mL),respectively,and processed by cold immersion and sonication for 30min.The peak ar-eas were compared by cold immersion and ultrasonication for 30 min.Ultrasonic extraction method was used to compare the peak areas of methanol solvent extracts at different time in-tervals.Results The results showed that the peak area of atractylosin,atractylosin,Eucalyptus al-cohol and Herba Palmatine was stable at different time points.The contents of essential oil such as atractylosin,Curcumol and eucalyptus alcohol increased obviously,while the content of atrac-tylosin decreased after processing.Conclusion The increased trend of atractylosin,curcutanol and Cineole in the volatile oil of Rhizoma atractylodis bran-stired might be related to the stron-ger spleen-strengthening effect and the weaker dryness of the Rhizoma atractylodis bran-stired.
