Effect of Shenshuaixiezhuo Decoction on Intestinal Microflora in Rats with Chronic Kidney Disease Complicated with Atherosclerosis
Objective To investigate the effect of Shenshuaixiezhuo Decoction on regulating in-testinal flora in chronic kidney disease(CKD)complicated with atherosclerosis(AS).Methods 30 SD rats were randomly divided into 3 groups,which were sham operation group,model group and Shenshuaixiezhuo Decoction group.Except sham operation group,the other groups were given 5/6 nephrectomy and high-fat diet to replicate CKD complicated with AS rat model.Shenshuaixiezhuo Decoction group was given 12.0 g/(kg·d)traditional Chinese medicine de-coction by intragastric administration,sham operation group and model group were given equal volume normal saline intragastric administration,which lasted for 8 weeks.The levels of blood creatinine(Scr),urea nitrogen(BUN),total cholesterol(TC),triglyceride(TG)were measured by automatic biochemical analyzer.HE staining was used to observe the histopathology of aorta.The intestinal flora structure of rats in each group was studied by 16S rDNA sequencing.Results Compared with model group,Scr,BUN,TC and TG levels in Shenshuaixiezhuo Decoction group were significantly decreased(P<0.01).Histological observation of aorta showed that the patho-logical changes of Shenshuaixiezhuo Decoction were less than that of model group.16S rDNA sequencing showed that Shenshuaixiezhuo Decoction could regulate the diversity and composi-tion of intestinal flora in CKD complicated with AS rats,and reduce the number of health disor-ders of the flora(P<0.05).Compared with the model group,Shenshuaixiezhuo Decoction in-creased the ratio of firmicutes to Bacteroides.The abundance of pathogenic bacteria Staphylococcus and Corynebacterium decreased,while the abundance of candidatus saccharimonas increased(P<0.05).LEfse analysis of the characteristic bacteria of Shenshuaixiezhuo Decoction group were Eu-bacterium mucosus,bifidobacterium,erysipelothrix,etc.Conclusion Shenshuaixiezhuo Decoction can interfere with the development of CKD As by regulating the composition of intestinal flora.
