Experimental Study on the Intervention of Insomnia after Stroke in Rats by Nourishing Heart and Tranquilizing Spirit Soup Combined with Auricular Pressure Beans
Objective To explore the mechanism and efficacy of Nourishing Heart and Tranquili-zing Spirit Soup combined with auricular acupoint pressure beans in intervening post-stroke in-somnia in rats.Methods A post-stroke insomnia model was constructed in rats,and after suc-cessful modeling,the rats were randomly divided into the sham-operation group,the model group,the western medicine group,and the experimental group,and intervened continuously for 14 days.The water maze experiment was conducted to record the evasion latency period of the rats before the treatment,and the elevated cross-maze experiment was conducted to record the number of times that the rats entered into the central area.After 2 weeks of treatment,the rats were treated and the expression of BDNF,MT,5-HT and NE in serum and hippocampal tis-sues of each group was measured by ELISA.The relative expression levels of NE transporter proteins and MT receptor proteins in rats of each group were detected by Western blot.Results The number of entries into the central zone,the expression levels of BDNF,MT in serum,and the expression levels of BDNF、MT、5-HT、NE、SLC6A3 and MTR1B proteins in the experimental group were higher than those in the model group(P<0.05).Conclusion Nourishing Heart and Tranquilizing Spirit Soup combined with auricular pressure bean has a certain improvement effect on the symptoms of post-stroke insomnia in rats,and its mechanism of action may be realized by elevating the 5-HT,BDNF in the body,and strengthening the expression of SLC6A3 and MTR1B proteins.
