目的 探究白细胞介素-9(IL-9)、白细胞介素-2 受体α亚单位(IL-2RA)和白细胞介素-2 受体β亚单位(IL-2RB)基因多态性在冠心病发生中的预测作用.方法 选取 2023 年 2 月-2024 年 2 月安阳市人民医院收治的冠心病患者 128 例,按 1∶1 选取同期体检健康者 128 例为对照组.比较冠心病组和对照组 IL-9 基因位点rs55692658、IL-2RA基因位点rs12569923、IL-2RB基因位点rs3218264 多态性频率差异.通过ROC曲线分析IL-9基因位点rs55692658、IL-2RA基因位点rs12569923、IL-2RB基因位点rs3218264 与联合数据预测冠心病发生的价值效能.结果 冠心病组IL-9 基因位点rs55692658 的AA基因型表达频率低于对照组(P<0.05).冠心病IL-2RA基因位点rs12569923 的CC基因型表达频率低于对照组(P<0.05).冠心病组IL-2RB基因位点rs3218264 的GG基因型表达频率低于对照组(P<0.05).ROC曲线分析显示,IL-9、IL-2RA和IL-2RB基因预测冠心病发生的AUC分别为 0.563、0.589、0.579;敏感度分别为 22.7%、49.2%、82.8%;特异度分别为 89.8%、68.7%、34.4%.联合数据预测冠心病发生的AUC为 0.661;敏感度为 55.5%;特异度为 68.7%.结论 冠心病易感性与IL-9 基因的位点rs55692658、IL-2RA基因的位点rs12569923、IL-2RB基因的位点rs3218264 多态性频率存在相关,IL-9、IL-2RA和IL-2RB基因多态性可作为临床预测冠心病发生风险生物学指标.
The Role of IL-9,IL-2RA,and IL-2RB Gene Polymorphisms in Coronary Heart Disease
Objective To explore the role of IL-9,IL-2RA,and IL-2RB gene polymorphisms in coronary heart disease.Methods 128 patients with coronary heart disease admitted to our hospi-tal from February 2023 to February 2024 were selected,and 128 healthy individuals who under-went physical examinations during the same period were selected as the control group in a 1∶1 ratio.The frequency differences of polymorphisms at sites rs12569926 of the IL-9 gene,rs12569923 of the IL-2RA gene,and rs3218264 of the IL-2RB gene between the coronary heart disease group and the control group was compared.And the ROC curve was used to analyze the value and efficacy of predicting the occurrence of coronary heart disease by analyzing the loci rs55692658 of the L-9 gene,rs12569923 of the IL-2RA gene,and rs3218264 of the IL-2RB gene in combination with data.Results The expression frequency of the A allele and AA geno-type of the IL-9 gene locus rs55692658 in the coronary heart disease group was lower than that in the control group(P<0.05).The expression frequency of the C allele and CC genotype of the IL-2RA gene locus rs12569923 in the coronary heart disease group was lower than that in the control group(P<0.05).The G allele and GG genotype expression frequency of the IL-2RB gene locus rs3218264 in the coronary heart disease group were lower than those in the control group(P<0.05).ROC curve analysis showed that the AUC of IL-9,IL-2RA,and IL-2RB genes predicting the occurrence of coronary heart disease were 0.563,0.589,and 0.579,respectively.The sensitivity values were 22.7%,49.2%,and 82.8%,respectively.The specificity was 89.8%,68.7%,and 34.4%,respectively.The IL-9,IL-2RA,and IL-2RB genes were included in the bina-ry logistic regression model to calculate the combined data.The joint data predicted the occur-rence of coronary heart disease with an AUC of 0.661,a sensitivity of 55.5%,and a specificity of 68.7%.Conclusion There is a correlation between susceptibility to coronary heart disease and the frequency of polymorphisms at sites rs55692658 of the IL-9 gene,rs12569923 of the IL-2RA gene,and rs3218264 of the IL-2RB gene.Polymorphisms in the IL-9,IL-2RA,and IL-2RB genes can serve as clinical biological indicators for predicting the risk of coronary heart disease.
