生物合成纳米硒治疗非酒精性脂肪肝病的机制研究

目的 研究生物合成纳米硒对小鼠非酒精性脂肪肝病(non-alcoholic fatty liver disease,NAFLD)的作用与机制.方法 C57BL/6J雄性小鼠 30 只,使用高脂、低蛋氨酸、胆碱缺乏(CDAHFD)饲料诱导小鼠NAFLD.小鼠随机分为 3 组:空白组(正常饲养 6 周)、模型组(CDAHFD饲料喂养 6 周,治疗组(CDAHFD饲料喂养 6 周+7.5 μg/20 g纳米硒灌胃6 周,每周1 次).记录小鼠体质量变化情况,实验周期结束后禁食12 h,解剖取材,检测血清肝功能、血脂指标;肝脏HE、油红O、马松染色观察肝脏病理学形态、脂质沉积及肝纤维化程度.qRT-PCR检测脂肪代谢、炎性细胞因子的基因表达水平.结果 与空白组相比,模型组小鼠谷丙转氨酶(ALT)、谷草转氨酶(AST)、甘油三酯(TG)、总胆红素(TB)显著增高(P<0.01),肝脏质量、肝脏系数显著升高(P<0.01),肝脏病理切片显示大量细胞肿胀变形、红色脂质沉积、胶原纤维生成,脂质代谢与炎症因子mRNA表达升高(P<0.05),与模型组比较,治疗组小鼠ALT、AST、TG、TB明显下降(P<0.05),肝脏质量、肝脏系数降低(P<0.05),肝脏病理切片提示细胞肿胀变形减少、脂质沉积降低、纤维化程度下降,脂质代谢与炎症因子mRNA表达下降(P<0.05).结论 生物合成纳米硒可有效改善高脂饮食诱导的小鼠NAFLD,减少脂质沉积、肝脏炎症与纤维化.
Mechanism of Biosynthesized Selenium-Nanoparticles in Treating Non-Alcoholic Fatty Liver Disease
Objective To investigate the effects and mechanisms of Bio-synthesized Selenium-nanoparticles on non-alcoholic fatty liver disease(NAFLD)in mice.Methods Thirty male C57BL/6J mice were used to induce NAFLD in mice using a high-fat,low-methionine,and choline-deficient(CDAHFD)diet.The mice were randomly divided into three groups:the blank group(Con,normal feeding for 6 weeks),the model group(CDAHFD,CDAHFD diet feeding for 6 weeks and PBS gavage),and the treatment group(CDAHFD+SeNPs,CDAHFD diet feed-ing for 6 weeks and 7.5 μg/20 g nano-selenium gavage once a week for 6 weeks).The weight changes of the mice were recorded.After the end of the experimental period,the mice were fasted for 12 hours,dissected for sample collection,and the serum liver function and lipid indica-tors were detected.The liver was stained with HE,oil red O,and Masson staining for observing the pathological morphology,lipid deposition,and the degree of liver fibrosis.qRT-PCR was uti-lized to detect the levels of fat metabolism and inflammatory cytokines.Results Compared with the Con group,the liver function indicators of ALT and AST in the model group were significant-ly increased(P<0.01),as were the blood lipid indicators of TG and TB(P<0.01).The liver weight and liver coefficient were also significantly increased(P<0.01).Histological sections of the liver showed a large number of cells swollen and deformed,red lipid deposits,and collagen fibers.The mRNA expression of lipid metabolism and inflammatory factors was increased(P<0.05).Compared with the model group,the liver function indicators of ALT and AST,as well as the blood lipid indicators of TG and TB,were significantly decreased(P<0.01)in the nano-sele-nium treatment group.The liver weight and liver coefficient were also significantly lower(P<0.01).Histological sections of the liver showed a reduction in cell swelling and deformation,a decrease in lipid deposits,and a lower degree of fibrosis.The mRNA expression of lipid metabo-lism and inflammatory factors was also decreased(P<0.05).Conclusions Biosynthesized nano-selenium can effectively ameliorate NAFLD in mice induced by a high-fat diet,reduce lipid dep-osition,liver inflammation and fibrosis.
