摘要

以黄瓜新品种宁运3号的种子为材料,进行组织培养和快速繁殖研究,比较了不同时间HgCl2溶液浸泡对种子萌发的影响,不同浓度的6-BA、NAA对不定芽及根诱导的影响.结果表明,用0.1 g100mLHgC12浸泡8 min,种子的萌发率较高,污染率较低;在0.1~1.0mg/L范围内随6-BA浓度的提高,不定芽诱导率提高,MS+6-BA 1.0 mg/L为较适宜的芽诱导培养基;1/2MS+NAA 0.5 mg/L对生根诱导效果较好.

Abstract

Using the seed of the new cucumber variety Ningyun No. 3 as material, tissue culture and rapid reproduction was studied. The effect of different immersing time in HgCl2 on seeds germination, influence of different concentrations of 6-BA, NAA on bud and root induction was studied. The results showed that immersed in 0.1 g/100mL HgCl2 for 8 min, relatively high germination ratio with low pollution rate could be achieved; In 0.1-1.0 mg/L, the induction rate of adventitious buds increased by the increasing of 6-BA's concentration; MS+1.0 mg/L 6-BA was the best medium for crowd bud propagation. The optimized medium for rooting was 1/2 MS+0.5 mg/L NAA.
