甘薯近缘野生种抗病基因同源序列的分离与鉴定

摘要
根据植物NBS-LRR类抗病基因同源序列(RGA)的保守区域设计简并引物,从甘薯近缘野生种Ipomoea grandifolia中扩增NBS-LRR类RGA,得到7条NBS-LRR类RGA,其编码的氨基酸序列包含NBS-LRR类RGA所具有的保守的P-loop、Kinase-2和Kinase-3及GLPL区.用DNAMAN软件对7个I.grandifolia NBS-LRR类RGA及10个已知物种的NBS-LRR类RGA氨基酸序列进行聚类分析,发现其聚为两类,两类之间的相似性较低,推测各类中相似性较高的序列分别属于同一个基因家族.
Abstract
The degenerate primers designed from the conserved motifs in the NBS-LRR resistance gene analog (RGA) of various plants were used to amplify NBS-LRR RGA from wild relative of Ipomoea batatas,7 NBS-LRR RGAs were obtained according to cloning and sequencing,of which the amino acid sequence contained P-loop,Kinase-2,Kinase-3 and GLPL motif of the NBS-LRR RGA.Cluster analysis of the predicted amino acid sequences grouped the 7 RGAs of I.grandifolia and the known 10 NBS-LRR RGAs in NCBI database into two classes,between which the sequence similarity was low.It was deduced that the RGAs with high similarity belonged to one gene family.
