Studies on Genomic DNA Extraction of Erythropsis kwangsiensis
Using the leaves of Erythropsis kwangsiensis as material,the genomic total DNA was extracted.Three methods of genomic total DNA extraction including improved CTAB method,reagent box method and improved SDS method were compared.Results showed that the DNA extracted from improved CTAB method and reagent box method had high quality and less impurity,but quantity of the latter was less than that of the former.The reagent box method had the advantages of simple operation,short time and less harmful,but more expensive than that of the improved CTAB method.In addition,improved SDS method didn't have the complete bands,so it was not suitable for genomic DNA extraction of Erythropsis kwangsiensis.The genomic DNA extracted from the leaves preserved in silica gel for 90 days were not significantly different from that of fresh leaves,and the yield of the former was much more than that of the latter.
