β-环糊精辅助提取茶末总黄酮的工艺优化

Optimizing β-CD Assisted Extraction of Total Flavonoids from Tea Dust

以制茶副产物茶末为原料,采用β-环糊精辅助提取茶末总黄酮,在单因素试验基础上,采用Box-Behnken试验设计和响应面分析法研究了料液比(g∶mL)、β-环糊精含量和提取温度对茶末总黄酮提取率的影响,并建立该工艺的二次多项式模型.结果表明,回归模型具有极显著性,可以对茶末总黄酮提取率进行很好地分析和预测;确定了茶末总黄酮提取工艺的最佳条件为料液比1∶61.83 (g∶mL)、β-环糊精含量8.27%、温度53℃,且在此条件下验证茶末总黄酮提取率为7.843%,接近预测值7.835%.与传统的水回流法和超微粉碎法相比,β-环糊精辅助提取法高效环保,且提取率更高.
The β-CD assisted extraction was used to extract total flavonoids from by-products of tea dust-tea.Based on the single factor experiment,the effects of material-to-solvent ratio,β-CD content and extraction temperature on the extraction efficiency of total flavonoids were investigated with the Box-Behnken experiment and the response surface analysis.A quadratic polynomial model was established and verified.The optimal conditions of extracting total flavonoids were solid-to-solvent ratio of 1∶61.83 (g∶ mL),β-CD content of 8.27%,extraction temperature of 53℃.Under these conditions,the extraction efficiency of total flavonoids was 7.843%,close to the predicted value of 7.835%.Compared with the traditional boiling reflux method and ultrafine grinding extraction technology,the 3-CD assisted extraction was efficient,environment friendly with high extraction rate.
