摘要

以果园土为菌种来源,采用平板分离法得到5种酵母菌,对其进行紫外线诱变,筛选出7株酵母菌突变株,通过测定突变株的发酵性能,从中筛选出1株液化酶活力、糖化酶活力、蛋白酶活力及发酵力均较好的酵母突变株N4-4,其发酵液化酶活力、糖化酶活力、蛋白酶活力和发酵力比对照菌株分别提高了6.7%、7.5%、8.1%和39.0%,通过单因素和正交试验确定该突变株的最佳发酵条件.结果表明,在30℃、120 r/rmin的培养条件下,突变株最佳发酵条件为3%葡萄糖、3% (NH4)2SO4、pH 5.0、3%接种量(V∶V).

Abstract

Five yeasts were isolated from orchard soil by streak plate method.By ultraviolet mutagenesis,seven mutant strains were screened.After examining the fermentation capacity of each mutant strain,a strain with high liquefied enzyme activity,sacharifying enzyme activity,protease activity and frementing power was screened and named as N4-4.Its fermentation liquefied enzyme activity,saccharifying enzyme activity,protease activity and fermenting power increased by 6.7%,7.5%,8.1% and 39.0% compared with the wild type strains.The optimal fermentation condition of this yeast was determined by single factor test and orthogonal test.The results showed that the optimal fermentation condition were 3% glucose,3%(NH4)2SO4,pH 5.0,3% inoculation amount (V∶V).
