Effects of ethyl acetate extract of Red ginseng on estrogen in rats with myocardial ischemia-reperfusion injury by regulating the PI3K/AKT/GSK3-β pathway
Objective To analyze the regulatory effect of ethyl acetate extract of Red ginseng on estrogen in myocardial ischemia-reperfusion injury rats through the phosphatidylinositol-3-OH kinase(PI3K)/protein kinase B(AKT)/glycogen synthase kinase 3-β(GSK3-β)signaling pathway.Methods Fifty female Sprague-Dawley(SD)rats in the specific pathogen-free(SPF)level were selected.Ten rats died during the modeling,and the remaining rats were randomly divided into blank group,model group,low-dose group,and high-dose group,with 10 rats per group.Rats in the control group were treated with blank control,and the remaining were subjected to the modeling of myocardial ischemia-reperfusion injury.Rats in low-dose group and high-dose group were further given ethyl acetate extract of red ginseng at the dose of 56.7mg/kg and 280mg/kg by intragastric administration,respectively.Myocardial damage(troponin I[cTnI],creatine kinase isoenzyme[CK-MB]),oxidative stress(malondialdehyde[MDA],superoxide dismutase[SOD],plasma glutathione peroxidase[GSH-Px]),nitric oxide(NO),inducible nitric oxide synthase(eNOS),sex hormones,and expression levels of key proteins in the PI3K/AKT/GSK3-β signaling pathway were detected.Results Compared with those of the blank group,the levels of cTnI,CK-MB,MDA,and estrogen(E2)in the model group,low-dose group,and high-dose group were all significantly higher,and the levels of SOD,GSH-Px,NO,eNOS,and protein expressions of PI3K,AKT and GSK3-β were significantly lower(all P<0.05).Compared with those of the model group,the levels of cTnI,CK-MB,MDA,and E2 in the low-dose group and the high-dose group were significantly lower,and the levels of SOD,GSH-Px,NO,eNOS,and protein expressions of PI3K,AKT and GSK3-β were significantly higher(all P<0.05).Compared with those of the low-dose group,the levels of cTnI and CK-MB and E2 in the high-dose group were significantly lower,and the levels of SOD,GSH-Px,NO,and eNOS and protein expressions of PI3K,AKT and GSK3-β expression were significantly higher(all P<0.05).Conclusion Ethyl acetate extract of Red ginseng can improve myocardial injury,inhibit oxidative stress and restore estrogen level in rats with myocardial ischemia-reperfusion injury by regulating the PI3K/AKT/GSK3-β signaling pathway.
