Correlation of serum lncRNA FAM138B and miR-105-5p expressions with the pathological features and prognosis in patients with non-small cell lung cancer
Objective To investigate the correlation of serum long non-coding RNA family with sequence similarity 138 member B(lncRNA FAM138B)and microRNA-105-5p(miR-105-5p)expressions with the pathological features and prognosis in patients with non-small cell lung cancer(NSCLC).Methods A total of 110 NSCLC patients admitted to our hospital from January 2018 to December 2019 were selected as the NSCLC group,and 60 healthy individuals receiving physical examination during the same period were selected as the control group.Serum lncRNA FAM138B and miR-105-5p expressions were detected by quantitative real-time polymerase chain reaction(qRT-PCR).The correlation of serum lncRNA FAM138B and miR-105-5p expressions with the pathological characteristics of NSCLC patients was analyzed.The target binding relationship between lncRNA FAM138B and miR-105-5p was predicted in the StarBase database.The correlation between serum lncRNA FAM138B and miR-105-5p expressions in NSCLC patients was analyzed by the Pearson correlation.Risk factors for the prognosis of NSCLC patients were analyzed by the multivariate Cox regression.Receiver operating characteristic(ROC)curves ere plotted to analyze the predictive value of serum lncRNA FAM138B and miR-105-5p expressions in the prognosis of NSCLC patients.Results Compared with those of the control group,serum lncRNA FAM138B expression was significantly downregulated and miR-105-5p expression was upregulated in the NSCLC group(P<0.05).LncRNAFAM138Bwas predicted by the StarBase database to have a complementary sequence to the promoter region of miR-105-5p.Pearson correlation analysis showed that serum lncRNA FAM138B was negatively correlated with miR-105-5p expression in NSCLC patients(r=-0.770,P<0.001).LncRNA FAM138B and miR-105-5p expressions were significantly correlated with the differentiation,tumor-node-metastasis(TNM)staging and lymph node metastasis of NSCLC(P<0.05).The 3-year overall survival of the 110 NSCLC patients was 56.36%(62/110).Kaplan-Meier survival curve analysis showed that the overall survival of NSCLC patients expressing high levels of lncRNA FAM138B was significantly higher than those expressing low levels.The overall survival of NSCLC patients expressing high levels of miR-105-5p was significantly higher than those expressing low levels(P<0.05).Multivariate Cox regression analysis showed that low differentiation,TNM staging III-IV,lymph node metastasis and miR-105-5p≥1.494 were independent risk factors for death in NSCLC patients,and lncRNA FAM138B≥0.871 was an independent protective factor(P<0.05).ROC curve analysis showed that the area under the curve(AUC)of serum lncRNA FAM138B and miR-105-5p expression alone and their combination in predicting the prognosis of NSCLC patients was 0.783,0.779 and 0.905 respectively.AUC of the combination detection was significantly larger than that of the single detection(P<0.05).Conclusion Low expression of serum lncRNA FAM138B and high expression of miR-105-5p in NSCLC patients are associated with the degree of differentiation,TNM staging,lymph node metastasis and prognosis in NSCLC patients,which may become auxiliary prognostic markers.
