The correlation of expression levels of lncRNA NCK1-AS1 and lncRNA TP73-AS1 in gastric cancer tissues with the Wnt/β-catenin signaling pathway and prognosis
Objective To study the correlation of expression levels of long non-coding ribonucleic acid(lncRNA)NCK1-AS1 and lncRNA TP73-AS1 in gastric cancer tissues with the Wnt/β-catenin signaling pathway and prognosis.Methods A total of 113 gastric cancer patients who underwent radical gastrectomy from February 2019 to October 2019 were selected.Their gastric cancer tissues and paracancerous tissues were taken during the surgery.The mRNA levels of lncRNA NCK1-AS1,lncRNA TP73-AS1 and Wnt3a,Wnt5a,β-catenin,cyclin D1,and proto-oncogene(c-myc)involved in the Wnt/β-catenin signaling pathway in gastric cancer tissues and paracancerous tissues were detected by real-time quantitative polymerase chain reaction(qRT-PCR).The correlation of expression levels of lncRNA NCK1-AS1 and lncRNA TP73-AS1 with the mRNA levels of key genes in the Wnt/β-catenin signaling pathway was analyzed by the Pearson correlation.The 3-year survival in gastric cancer patients was analyzed by plotting the Kaplan-Meier survival curves.Results The expression levels of lncRNA NCK1-AS1 and lncRNA TP73-AS1 in gastric cancer tissues were significantly higher than those of paracancerous tissues(P<0.05).The mRNA levels of Wnt3a,Wnt5a,β-catenin,cyclin D1 and c-myc in gastric cancer tissues were significantly higher than those of paracancerous tissues(P<0.05).Pearson correlation analysis showed that the mRNA levels of Wnt3a,Wnt5a,β-catenin,cyclin D1 and c-myc in the Wnt/β-catenin signaling pathway were positively correlated with those of lncRNA NCK1-AS1 and lncRNA TP73-AS1(P<0.05).The expression levels of lncRNA NCK1-AS1 and lncRNA TP73-AS1 were significantly correlated with the tumor-node-metastasis(TNM)staging,infiltration depth,and lymph node metastasis in gastric cancer patients(P<0.05).Patients were followed up for 3 years,with the median follow-up time of 27.90 months.The Kaplan-Meier survival curves showed that the 3-year survival in gastric cancer patients expressing high levels of lncRNA NCK1-AS1 was significantly lower than those expressing low levels(52.31%[34/65]vs 77.08%[37/48],P<0.05).The 3-year survival in gastric cancer patients expressing high levels of lncRNA TP73-AS1 high expression group was significantly lower than those expressing low levels(52.78%[38/72]vs 80.49%[33/41],P<0.05).Conclusion LncRNA NCK1-AS1 and lncRNA TP73-AS1 are upregulated in gastric cancer tissues,which may promote the occurrence and development of gastric cancer by activating the Wnt/β-catenin signaling pathway and lead to poor prognosis.They are expected to serve as the biomarkers for evaluating the prognosis of gastric cancer.
