Effect of total flavonoids extracted from Kudiezi on vascular endothelial function in angina pectoris rats with coronary heart disease based on the PI3K/AKT signaling pathway
Objective To explore the effect of total flavonoids extracted from Kudiezi on vascular endothelial function in angina pectoris rats with coronary heart disease based on the phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)signaling pathway.Methods A total of 85 Sprague-Dawley(SD)rats in the specific pathogen free(SPF)level were used,with 20 in the control group,and 65 for the modeling of angina pectoris-coronary heart disease.Finally,60 successfully modelling rats were randomly divided into model group,low-dose total flavonoid group and high-dose total flavonoid group,with 20 per group.Gavage of 4mL of normal saline was performed per rat in control group and model group.In addition,4mL of normal saline containing 1.0mg/mL and 2.0mg/mL total flavonoids extracted from Kudiezi was administrated by gavage in rats of low-dose and high-dose total flavonoid groups,respectively.The vascular endothelial function,myocardial enzyme levels,inflammatory factor levels and protein expressions of key proteins in the PI3K/AKT signaling pathway were compared.Results Compared with those of control group,rats in the model group and low-dose total flavonoid group presented significantly higher expression levels of endothelin-1(ET-1),creatine kinase-MB(CK-MB),lactate dehydrogenase(LD),tumor necrosis factor α(TNF-α)and interleukin-6(IL-6),and significantly lower expression levels of NO,PI3K and AKT(P<0.05).Compared with those of model group,rats in the low-dose and high-dose total flavonoid groups presented significantly lower levels of ET-1,CK-MB,LD,TNF-α and IL-6,and significantly higher expression levels of NO,PI3K and AKT(P<0.05).Compared with those of low-dose total flavonoid group,rats in the high-dose total flavonoid group presented significantly lower levels of ET-1,CK-MB,LD,TNF-α and IL-6,and significantly higher expression levels of NO,PI3K and AKT(P<0.05).Conclusion The total flavonoids extracted from Kudiezi can effectively regulate vascular endothelial function,improve myocardial enzyme levels,inhibit the expression of inflammatory factors and relieve symptoms in angina pectoris rats with coronary heart disease by promoting the PI3K/AKT signaling pathway.
