Expression of lncRNA SNHG18 in bladder cancer and its effect on its malignant biological behaviors
Objective To detect the expression of long non-coding RNA(lncRNA)small nucleolar RNA host gene 18(SNHG18)in bladder cancer cell lines,and to analyze the correlation of it with the malignant biological behaviors.Methods A total of 42 patients with bladder cancer who underwent surgery from August 2021 to October 2022 were selected.Reverse transcription quantitative real-time polymerase chain reaction(RT-qPCR)was used to detect the expression of lncRNA SNHG18 in 42 bladder cancer tissues and 3 bladder cancer cell lines(5637,T24,SW780).Bladder cancer cells were transfected with the constructed overexpression vector pcDNA3.1-SNHG18 and knock-down vector si-SNHG18.Cell proliferation assay and colony formation assay were performed to detect cell proliferative ability.Scratch test and Transwell assay were used to detect cell migration and invasion.Results LncRNA SNHG18 was significantly downregulated in bladder cancer tissues and cells than that of normal bladder epithelial tissues and bladder epithelial cells(P<0.05).No correlation was found between the expression of lncRNA SNHG18 and sex,age,smoking,hypertension,diabetes,lymph node and distant metastasis and clinical stage of bladder cancer patietns(P>0.05).Overexpression of SNHG18 inhibited the proliferation,migration and invasion of bladder cancer cells in vitro(P<0.05).Knockdown of SNHG18 enhanced the proliferation,migration and invasion of bladder cancer cells in vitro(P<0.05).Conclusion LncRNA SNHG18 is downregulated in bladder cancer tissue and it is not correlated with clinical parameters of bladder cancer patients.However,it is able to regulate the proliferation,migration and invasion of bladder cancer cells.
