目的 探讨微小RNA-34a-5p(miR-34a-5p)、高迁移率族蛋白B1(HMGB1)在宫颈癌(CC)及癌前病变患者表达水平及二者与人乳头状瘤病毒(HPV)感染的关系.方法 回顾性分析2018年1月至2022年6月诊治的165例宫颈手术标本,其中CC 55例(CC组),癌前病变55例为癌前病变组,正常宫颈组织55例为对照组.检测所有受试者宫颈组织miR-34a-5p、HMGB1表达水平及HPV感染情况;分析CC及宫颈癌前病变患者宫颈组织miR-34a-5p、HMGB1表达水平与HPV感染的关系;生物信息学预测miR-34a-5p与HMGB1的关系,分析CC及宫颈癌前病变患者宫颈组织miR-34a-5p与HMGB1水平的相关性.结果 CC组宫颈组织miR-34a-5p表达水平低于对照组和癌前病变组(P<0.05),HMGB1表达水平、HPV阳性感染率高于对照组和癌前病变组(P<0.05);癌前病变组宫颈组织miR-34a-5p表达水平低于对照组(P<0.05),HMGB1表达水平、HPV阳性感染率高于对照组(P<0.05).宫颈组织miR-34a-5p表达水平与CC及其癌前病变患者HPV感染呈负相关(P<0.05);宫颈组织HMGB1表达水平与CC及癌前病变患者HPV感染呈正相关(P<0.05).miR-34a-5p与HMGB1存在结合位点,癌前病变、CC患者宫颈组织miR-34a-5p表达水平均与HMGB1呈负相关(P<0.05).结论 CC及癌前病变患者宫颈组织miR-34a-5p表达下调,HMGB1表达上调,二者均与HPV感染密切相关.
Expression levels of miR-34a-5p and HMGB1 in cervical cancers and precancerous lesions of cervical tissue and their correlation with HPV infection
Objective To investigate the expression levels of microRNA-34a-5p(miR-34a-5p)and high mobility group box protein B1(HMGB1)in cervical cancer(CC)and precancerous lesions and their relationship with human papilloma virus(HPV)infection.Methods A total of 165 cervical surgical specimens in our hospital from January 2018 to June 2022 were retrospectively analyzed,including 55 CC specimens,55 precancerous lesions and 55 normal cervical tissues.The expression levels of miR-34a-5p and HMGB1 in clinical specimens,and HPV infection in all subjects were detected.The correlation of the expression levels of miR-34a-5p and HMGB1 in cervical tissue with HPV infection in patients with CC and cervical precancerous lesions was analyzed.The relationship between miR-34a-5p and HMGB1 was predicted by bioinformatics,and the correlation of expression level of miR-34a-5p with that of HMGB1 in cervical tissue of patients with CC and cervical precancerous lesions was analyzed.Results The expression level of miR-34a-5p in CC tissues was significantly lower than that of normal cervical tissues and precancerous lesions(P<0.05),while the expression level of HMGB1 and the positive infection rate of HPV were higher(P<0.05).The expression level of miR-34a-5p in cervical precancerous lesions was significantly lower than that of normal cervical tissues(P<0.05),while the expression level of HMGB1 and the positive infection rate of HPV were significantly higher(P<0.05).The expression level of miR-34a-5p in cervical tissue was negatively correlated with HPV infection in patients with CC and precancerous lesions(P<0.05).The expression level of HMGB1 in cervical tissue was positively correlated with HPV infection in patients with CC and precancerous lesions(P<0.05).MiR-34a-5p had a binding site with HMGB1,and the expression level of miR-34a-5p in precancerous lesions of CC and CC specimens was negatively correlated with that of HMGB1(P<0.05).Conclusion MiR-34a-5p is down-regulated and HMGB1 is up-regulated in CC and precancerous lesions,both of which are closely related to HPV infection.
