Expression levels of Chemerin and TNF-α in peripheral blood,cord blood and umbilical cord tissue of gestational diabetes mellitus
Objective To measure the expression levels of Chemerin and tumour necrosis factor alpha(TNF-α)in mothers with gestational diabetes mellitus(GDM)and the fetuses,and to clear the effect of Chemerin and TNF-α on the pathogeny of GDM and development of fetuses.Methods Fifty-three pregnant women with GDM who received prenatal examination and delivered from August 2016 to November 2017 were selected as the GDM group,and 59 healthy pregnant women who received prenatal examination and delivered during the same period selected as the normal pregnancy group(NGT group).Serum Chemerin and TNF-α levels in peripheral blood of pregnant women and cord blood were determined by enzyme-linked immunosorbent assay(ELISA).The mRNA and protein expressions of Chemerin and TNF-α in umbilical cord were determined by real-time quantitative PCR and Western blot,respectively.Results There was no significant difference in serum Chemerin level between GDM group and NGT group(P>0.05),while Chemerin level in cord blood was significantly higher than that of NGT group(P<0.01).Chemerin level in maternal blood was significantly lower than that of umbilical cord blood in both GDM group and NGT group(P<0.01).Compared with that of NGT group,serum TNF-α level in GDM group was significantly higher(P<0.01).There was no significant difference in cord blood TNF-α level between GDM group and NGT group(P>0.05).Maternal blood TNF-α level in the GDM group was significantly higher than that of cord blood level(P<0.01),but there was no significant difference between them in the NGT group(P>0.05).The mRNA expression of Chemerin in the GDM group was significantly higher than that of NGT group(P<0.05).The mRNA expression of TNF-α in the GDM group was significantly higher than that of NGT group(P<0.05).The protein expression of Chemerin was similar between the two groups(P>0.05).The protein expression of TNF-α in the GDM group was significantly higher than that of NGT group(P<0.05).The correlation analysis showed a negative correlation between cord blood levels of Chemerin and TNF-α in the GDM group(P<0.05).In both GDM group and NGT group,the mRNA and protein expressions of TNF-α were positively correlated with neonatal body weight(P<0.05).Conclusion Fetal development is regulated by intrauterine inflammation,and cord blood Chemerin in GDM patients can play an indirect role through TNF-α.
