目的 研究HCG22在皮肤鳞癌(SSCC)患者血清中的表达水平,探讨其对宫颈癌诊断及高危HPV感染预测的临床价值.方法 纳入2018年12月至2021年2月收治的125例SSCC患者,其中包括78例高危型HPV(HR-HPV)患者,同时选取76例健康人群作为对照.RT-qPCR检测SSCC患者和对照受试者血清中HCG22的表达,ROC曲线分析HCG22对SSCC的诊断效能.根据SSCC患者血清中HCG22表达水平的中位值将SSCC患者分为HCG22低表达组和高表达组,分析其与SSCC患者临床指标的相关性.分析HCG22与SSCC患者的病理指标的相关性.Logistics多因素回归分析HCG22是否与高危型HPV感染SSCC具有独立相关性.结果 和健康受试者相比,SSCC患者血清中HCG22表达显著降低,其诊断SSCC的ROC曲线下面积(area under the curve,AUC)为0.9394,截点值为0.3850,灵敏度88.18％,特异度92.00％.HR-HPV阳性SSCC患者血清HCG22表达显著高于HR-HPV阴性SSCC患者,HCG22诊断SSCC为HR-HPV阳性的ROC曲线下面积为0.7381,截点值为0.3150,灵敏度83.33％,特异度68.09％.HCG22表达水平与临床分期、肿瘤分化程度、淋巴结转移、浸润肌层深度、高危型HPV(HR-HPV)感染显著相关(P＜0.05),且HCG22与皮肤鳞癌高危型HPV感染具有独立相关性(P＜0.05).结论 HCG22在高危型HPV感染皮肤鳞癌患者血清中低表达,其与皮肤鳞癌高危型HPV感染具有独立相关性,对皮肤鳞癌高危型HPV感染具有一定的辅助诊断价值.
Expression and clinical significance of LncRNA HCG22 in skin squamous cell carcinoma tissues and cells
Objective This article aims to study the serum expression of human leukocyte antigen complex group 22(HCG22)of patients with skin squamous cell carcinoma(SSCC),and to explore its clinical value for the diagnosis of cervical cancer(CC)and the prediction of high-risk human papillomavirus(HR-HPV)infection.Methods A total of 125 patients with SSCC admitted from December 2018 to February 2021 were enrolled in this study,including 78 patients with HR-HPV.In the meantime,76 healthy people were selected as controls.The serum expression of HCG22 in the subjects was detected by real-time quantitative reverse transcription polymerase chain reaction(RT-qPCR),and the diagnostic efficacy of HCG22 in SSCC was analyzed by receiver operating characteristic(ROC)curve.SSCC patients were divided into HCG22 low expression group and high expression group based on the median value of HCG22 serum expression.The association between HCG22 serum expression and clinical indicators,between HCG22 serum expression and pathological indexes in SSCC patients was analyzed.Logistics multivariate regression was used to analyze whether HCG22 was independently associated with HR-HPV infection in SSCC.Results Compared with healthy subjects,the serum expression of HCG22 in SSCC patients was significantly lower,the area under the ROC curve(AUC)for diagnosing SSCC was 0.9394,with 0.3850 of the cut-off value,88.18％of the sensitivity,and 92.00％of the specificity.The serum expression of HCG22 in SSCC patients with positive HR-HPV was significantly higher than that of SSCC patients with negative HR-HPV.The AUC of HCG22 for diagnosing SSCC in HR-HPV positive,cutoff value,sensitivity,and specificity was 0.7381,0.3150,83.33％,and 68.09％,respectively.The serum expression of HCG22 was significantly correlated with clinical stage,tumor differentiation,lymph node metastasis,depth of muscle-invasion,and HR-HPV infection,and HCG22 was independently correlated with HR-HPV infection in SSCC.Conclusion HCG22 is lowly serum expression in the SSCC patients with HR-HPV,which is independently correlated with HR-HPV infection in SSCC,with a certain auxiliary diagnostic value for HR-HPV infection in SSCC
