Box-Behnken设计-响应面法优化芦笋粉中黄酮的提取工艺

采用低共熔溶剂法从芦笋粉末中提取黄酮,通过Box-Behnken设计-响应面法对提取工艺进行了优化.结果表明:低共熔溶剂氢键供体与受体的选择以及氢键供体和受体的比例,对黄酮的提取有较为显著的影响,其中氢键供体选择尿素,受体选择氯化胆碱,供体受体比例为1∶2对黄酮的提取效率最佳,此外提取工艺的3个因素,提取时间、提取温度、料液比对黄酮的提取效率也有一定的影响,其影响程度按照由大到小的次序为:料液比、提取时间、提取温度.提取芦笋粉中黄酮的最优条件为:料液比1∶2,提取时间2 h,提取温度50 ℃.此工艺条件下得到总黄酮提取率为25.99%,响应面模型预测值为21.63%.
Optimization of Extraction Process of Flavonoids from Asparagus Powder with Box-Behnken Design-response Surface Method
The eutectic solvent method was used to extract flavonoids from asparagus powder,and the extrac-tion process was optimized by the Box-Behnken design-response surface method.The results showed that the selection of hydrogen bond donor and acceptor and the ratio of hydrogen bond donor and acceptor of low eutec-tic solvent had a significant impact on the extraction of flavonoids,and the extraction efficiency of flavonoids was the best when urea was selected as hydrogen bond donor,choline chloride as acceptor,and the ratio of donor to acceptor was 1∶2.The optimal conditions for the extraction of flavonoids from asparagus powder were as follows:solid-liquid ratio of 1∶2,extraction time of 2 h,and extraction temperature of 50 ℃.Under the conditions of this process,the extraction rate of total flavonoids was 25.99%,and the predicted value of the response surface model was 21.63%.
