Pathogen Isolation and Identification of Cyprinus carpio haematopterus Skin Ulcer Disease and Sensitivity Test of Chinese Herbal Medicines
The pathogenic bacteria causing skin ulcer of Cyprinus carpio haematopterus were determined and effective Chinese herbal medicines were obtained.The animal infection test confirmed that Shewanella putrefaciens,a Gram-negative bacterium named CC-1 that was isolated from the ulcer tissue of the diseased K.carp was the pathogenic bacterium through gram staining,physiological and biochemical identification,16Sr DNA sequence analysis,pathogenicity detection,and Chinese herbal medicine sensitivity test.The Chinese herbal medicine sensitivity test showed that S.putrefaciens was highly sensitive to Galla chinensis,Sappan lignum,and Coptidis rhizome,moderately sensitive to Astragalus membranaceus,Anemarrhena asphodeloides Bunge,and Rhizoma fagopyri cymosi,and insensitive to Poria cocos and Polygonum cuspidatum.The MIC(minimum inhibitory concentration)and MBC(minimum bactericidal concentration)of 13 Chinese herbs with antibacterial effects were determined.The results showed that the MIC and MBC of S.lignum,Scutellariae radix,Fructus mume,and G.chinensis were 7.81 mg/mL and 15.63 mg/mL respectively,followed by C.rhizoma with the MIC and MBC of 15.63 mg/mL and 31.25 mg/mL respectively.
