化浊解毒颗粒对脂多糖所致的急性肺损伤大鼠炎症相关指标的影响

Effect of Huazhuo Jiedu Granules on Inflammation Related Indicators in Rats with Acute Lung Injury Caused by Lipopolysaccharide

目的:探讨化浊解毒颗粒对急性肺损伤(Acute lung injury,ALI)大鼠炎症相关指标的影响.方法:60 只大鼠随机分为空白组,模型组,泼尼松组,化浊解毒低、中、高剂量组.除空白组外,其余各组给药 7d后构建ALI模型.行肺组织病理学观察及肺损伤评分,测定肺组织湿/干重比(W/D)、肺通透性指数、动脉血氧分压(PaO2)、肺泡灌洗液(BALF)炎性细胞分类计数,肺组织核因子κB(NF-κB)p65、肿瘤坏死因子α(TNF-α)、白细胞介素 6(IL-6)、白细胞介素 10(IL-10)mRNA表达,并检测肺组织白细胞介素 1β(IL-1β)、巨噬细胞炎性蛋白 1a(MIP-1a)含量.结果:与空白组比较,模型组大鼠肺组织结构紊乱,间质水肿,炎性细胞浸润;同时肺损伤评分、肺组织W/D、肺通透性指数、BALF炎性细胞数目,肺组织NF-κB p65、TNF-α、IL-6 mRNA表达,IL-1β、MIP-1a水平均升高(P<0.05),PaO2、IL-10 mRNA表达均降低(P<0.05).与模型组相比,化浊解毒低、中、高剂量组肺组织炎性细胞浸润及水肿不同程度减轻,且肺损伤评分、肺组织W/D、肺通透性指数、BALF炎性细胞数目、肺组织NF-κB p65 mRNA表达、IL-1β、MIP-1a水平均降低(P<0.05),PaO2升高(P<0.05);化浊解毒中、高剂量组肺组织IL-6 和TNF-α mRNA表达降低(P<0.05),IL-10 mRNA表达升高(P<0.05).结论:化浊解毒颗粒能改善ALI模型大鼠肺组织病理损伤,其机制可能与调控肺内炎症因子水平有关.
Objective:To explore the effect of Huazhuo Jiedu Granules(granules for eliminating turbid and removing toxin)on in-flammation related indicators in rats with acute lung injury(ALI).Methods:Sixty rats were divided into blank group,model group,prednisone group,low-,medium-,and high-dose groups of Huazhuo Jiedu Granules.After 7 days of administration,ALI model rats were constructed.Observation of lung tissue pathology and lung injury score were used to measure lung tissue wet/dry weight ratio(W/D),lung permeability index,arterial partial pressure of oxygen(PaO2),classification count of inflammatory cells in bronchoalveolar lavage fluid(BALF),lung tissue nuclear factor κ B(NF-κ B)P65,Tumor Necrosis Factor α(TNF-α),mRNA expression of inter-leukin-6(IL-6)and interleukin-10(IL-10),and to detect interleukin-1 β(IL-1β)in lung tissue and macrophage inflammatory pro-tein-1a(MIP-1a).Results:Compared with the blank group,the lung tissue structure of the model group was disordered,with intersti-tial edema and inflammatory cell infiltration;At the same time,lung injury score,lung tissue W/D,lung permeability index,BALF inflammatory cell count increased(P<0.05),PaO2 decreased(P<0.05),and NF-κ B p65,TNF-α,IL-6 mRNA expression in-creased(P<0.05),IL-10 mRNA expression decreased(P<0.05),IL-1 β and MIP-1a increased(P<0.05).Compared with the model group,the low-,medium-,and high-dose groups of Huazhuo Jiedu Granules showed that inflammatory cell infiltration and edema in lung tissue in varying degrees,and lung injury score,lung tissue W/D,lung permeability index,and BALF inflammatory cell count decreased(P<0.05),PaO2(P<0.05)increased,and mRNA expression of NF-κ B p65 in lung tissue reduced(P<0.05),IL-1 β and MIP-1a decreased(P<0.05);mRNA expression of IL-6 and TNF-α Medium-and high-dose groups decreased(P<0.05),while IL-10 mRNA expression increased(P<0.05).Conclusion:Huazhuo Jiedu Granules can improve the pathological damage of lung tissue in ALI model rats,and its mechanism may be related to regulating the levels of inflammatory factors in the lungs.
