Study on Anatomical Observation and Endogenous Hormone Regulation of Rooting on Gleditsia sinensis Softwood Cutting
To study the characteristics of rooting anatomical structure,and to investigate and analyze the dy-namic changes and effects of endogenous hormones in the process of adventitious root development during soft-wood cuttings of Gleditsia sinensis,so as to provide reference for further research on the development mecha-nism and regulation techniques of G.sinensis.The fresh branches of G.sinensis'Yuzao No.2'were used as the experimental material,the changes of morphological and structural characteristics of softwood cuttings during the development of adventitious roots were observed,the contents of IAA,ABA,GA3,and ZR of G.sinensis were determined by using HPLC.The results showed that the adventitious root primordia of the softwood cutting root-ing of G.sinensis developed from the cambium directly opposite the pith ray,which formed by mutagenic root primordium growth.The rooting process of G.sinensis was divided into three main stages:callus induction stage(0-20 d after cutting),adventitious root induction stage(20-40 d after cutting)and adventitious root expres-sion and extension stage(40 d after most cuttings adventitious root stretch out).In the critical period of ad-ventitious root formation,the increase of IAA content is the key to induce adventitious root primordium.Com-pared with the control group,the IAA content of ABT1 treatment group reached the peak at 30 days,which was 63.79 ng/g(FW).The ratio of IAA/ABA and IAA/ZR was also the highest.The change trend of ABA,GA3 and ZR was opposite to that of IAA.The adventitious root formation of G.sinensis belongs to the type of induce-to-root.During the critical pe riod of adventitious root formation,the increase in IAA content is the key to inducing root primordium.The ap-plication of ABT1 not only increases the endogenous hormone IAA content that is conducive to rooting on G.sinensis cuttings,but also promotes the dynamic balance of various endogenous hormones contents,inducing the occurrence of adventitious roots.
