Screening of a Phosphate-solubilizing Bacillus Strain and Evaluation of Its Growth Promoting Effect
Screening microbial strains that can effectively dissolve insoluble phosphorus can provide strain re-sources for the development of high utilization phosphorus fertilizer products.The strains that can dissolve both inorganic and organic phosphorus were isolated and purified through gradient dilution,and their phosphorus sol-ubilization ability was quantitatively detected through selective medium screening.Using a bacterial phylogenetic tree constructed based on the 16S rDNA gene sequence,the taxonomic status of the strain was determined.The IAA,protease,cellulase,and ferritin production capacity was conducted to elucidate the mechanism of bacterial growth promotion.The growth promoting effect of the strain was verified through a potted experiment of corn seedlings.The results showed that one strain of Lysinibacillus sphaericus SY50 was screened,which can dis-solve insoluble phosphorus.Its ability to dissolve inorganic phosphorus and organic phosphorus reached 386.51 and 101.42 mg/L,respectively.It can also produce various plant growth promoting substances such as IAA,protease,cellulase,and ferritin,with a good growth promoting effect on corn seedlings.L.sphaericus SY50 is a growth promoting strain with phosphorus solubilizing ability,providing excellent strain resources for the devel-opment of phosphorus solubilizing microbial agents.
