中药单体治疗脊髓损伤具有多途径、多靶点、不良反应少等优势,主要是通过调控Janus激酶/信号转导和转录激活因子(Janus kinase/signal transducer and activator of transcription,JAK/STAT)、核因子-κB(nuclear factor-κB,NF-κB)、核因子 E2相关因子 2/血红素加氧酶-1(nuclear factor E2 related factor/heme oxygenase-1,Nrf2/HO-1)、丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)、磷脂酰肌醇 3-激酶/蛋白激酶 B(phoinositide 3-kinase/protein kinase B,PI3K/Akt)、Wnt/β-catenin等信号通路,发挥降低炎症因子表达、抑制氧化应激反应、促进轴突再生、减少细胞凋亡等作用,从而修复脊髓损伤。但现有研究仍存在一些不足,如中药单体对脊髓损伤的有效性缺乏临床试验数据佐证,基础实验中脊髓损伤模型病证单一,信号通路及靶点上下游通路蛋白间的相互作用机制尚未阐明等,今后可借助分子生物技术、多组学技术、网络药理学等进一步明确中药单体修复脊髓损伤的作用机制,同时挖掘中医药更多潜在的分子标志物,为新药研发提供靶点。
Research Progress of Traditional Chinese Medicine Monomer Regulating Signal Pathways to Treat Spinal Cord Injury
Herbal monotherapy for spinal cord injury has the advantages of multi-pathway,multi-target and less adverse effects,mainly through the regulation of Janus kinase/signal transducer and activator of transcription(JAK/STAT),nuclear transcription factor-(JAK/STAT),nuclear factor-κB(NF-κB),nuclear factor E2 related factor/heme oxygenase-1(Nrf2/HO-1),mitogen-activa-ted protein kinase(MAP),and other activators of transcription(MAP).activated protein kinase(MAPK),phosphatidylinositide 3-kinase/protein kinase B(PI3K/Akt),Wnt/β-catenin and other signaling pathways,which play the role of reduce the expression of in-flammatory factors,inhibit oxidative stress,promote axonal regeneration,and reduce apoptosis,to repair spinal cord injury.However,there are still some shortcomings in the existing studies,such as the lack of clinical data to support the effectiveness of TCM monomers on spinal cord injury,a single spinal cord injury model in the basic experiments,the signaling pathway and the target upstream and downstream interactions between the pathway proteins have not yet been elucidated,and so on.In the future,we can use molecular bio-technology,multi-histology technology,network pharmacology,etc.,to further define the mechanism of the role of the TCM monomers in the repair of spinal cord injury,and at the same time,we can explore more potential molecular mechanisms of Chinese medicine.In the future,we can use molecular biotechnology,multi-omics technology and network pharmacology to further clarify the mechanism of action of TCM monomers in repairing spinal cord injury,and at the same time,we can explore more potential molecular markers of TCM,which can provide targets for the development of new drugs.
