探讨IL-34在小鼠慢性根尖周炎模型中的动态表达

目的:探讨白介素-34(IL-34)在小鼠慢性根尖周炎模型中的表达及变化情况.方法:选择35 只SD小鼠,对其下颌第一磨牙开髓制造根尖周炎的模型,并在0、7、14、21、28 d随机脱颈处死,解剖分离含有第一磨牙的下颌骨,进行影像学检查,了解模型中根尖周的破坏情况.将下颌骨标本制备切片,进行HE染色观察根尖周组织病理学改变以及抗酒石酸酸性磷酸酶(TRAP)染色检测破骨细胞,实时荧光定量PCR(RT-qPCR)检测IL-34 与肿瘤坏死因子-α(TNF-α)在根尖周炎模型中的动态表达.结果:影像学检查显示,0~7 d根尖无明显异常,14~21 d开始出现轻度根尖破坏,28 d根尖阴影变大,范围增宽.HE染色显示,0 d根尖周组织正常未见明显改变,7 d炎性因子浸润开始增多,14 d出现明显的骨质破坏以及坏死中心,21 d炎性渗出减少,小范围的骨质破坏区域增多,28 d炎性细胞浸润趋于稳定,小范围的骨质破坏连接成片,大范围的骨质破坏形成.TRAP染色显示,0 d几乎看不到破骨细胞,7~14 d破骨细胞数目明显增加,在21d时到达高峰,28 d时轻微下降,但7、14、21、28d时均高于0d(P<0.05).RT-qPCR检测显示,7~28 d的IL-34 及TNF-α的mRNA表达量均逐渐增高,且在14d到达顶峰,在21d呈现下降的趋势,28 d又呈现上升趋势,在7~28d的表达量均高于 0d(P<0.05),并且呈现正相关关系.结论:IL-34 的mRNA表达量随着根尖周炎病变程度发展而改变,尤其是与骨质破坏的程度相关,并与TNF-α有着密切的联系,出现短暂的表达量下降说明其表达受到了抑制,有助于为临床治疗提供方向.
Exploring dynamic expression of IL-34 in mouse model of chronic apical periodontitis
Objective:To explore the expression and changes of IL-34 in mouse model of chronic apical periodontitis.Methods:35 SD mice were selected to create a model of apical periodontitis by pulp opening of their mandibular first molar,and they were randomly decapitated and killed on days 0,7,14,21,and 28.The mandible containing the first molar was anatomically separated and X-ray imaging examinations were conductd to observe the periapical damage in the model.The mandibular specimens were prepared for sectio-ning,and HE staining was used to observe the histopathology changes around the root tip,and tartrate resistant acid phosphatase(TRAP)staining was used to detect osteoclasts.The dynamic expression of IL-34 and TNF-α in a periapical periodontitis model were detecetd by real-time fluorescence quantitative polymerase chain reaction(RT-qPCR).Results:X-ray examination indicated that there was no obvious abnormalities in the apical area from 0 to 7d;mild apical damage began to appear from 14d to 21d;on 28d,the root tip shadow became larger and the range widened.HE staining showed that the periapical tissue was normal and no significant changes were observed on 0d;the infiltration of inflammatory factors began to increase on 7d;on 14d,obvious bone destruction and necrotic centers appeared;on 21d,there was a decrease in inflammatory exudation and an increase in small areas of bone destruction;after 28d,the infiltration of inflammatory cells tended to stabilize,with small-scale bone destruction connecting into patches,and large-scale bone de-struction forming.TRAP staining showed that there was almost no osteoclast on 0d,the number of osteoclast increased significantly from 7d to 14d,reached the peak on 21d,and slightly decreased on 28d,but was higher than 0d on 7d,14d,21d,and 28d(P<0.05).RT qPCR detection showed that the mRNA expression levels of IL-34 and TNF-α from 7d to 28d were all gradually increased,reaching their peak on 14d,showing a decreasing trend on 21d,and then showing an upward trend on 28d.And the expression levels of both from 7d to 28d were higher than those of 0d(P<0.05),showing a positive correlation.Conclusion:The mRNA expression level of IL-34 changes with the development of periapical inflammation,especially in relation to the degree of bone destruction and TNF-α.There is a close connection,and a brief decrease in expression indicates that its expression has been suppressed,which provides direc-tion for our clinical treatment.
