Effect of fucoxanthin on viability and apoptosis of human cervical cancer HeLa cells
Objective:To investigate the effect of fucoxanthin on proliferation and apoptosis of HeLa cells and to analyze its mecha-nism of inhibiting tumor growth.Methods:A comparison was conducted between a treatment group of cervical cancer HeLa cells with fucoxanthin and a blank control group.MTT method was used to detect HeLa cells treated with different concentrations of fucoxanthin for 24 hours and the changes in the survival rate of HeLa cells after 12 hours,24 hours,and 48 hours of treatment with 0.5 umol/L of fucoxanthin were observed.The apoptosis of HeLa cells was detected using Annexin V/PI flow cytometry at 24 hours and 48 hours after stimulation with 0.1umol/L and 0.5umol/L of fucoxanthin,respectively.The production of ATP and total oxygen uptake were deter-mined after the culture medium of HeLa cells were stimulated with 0.1 umol/L,0.5 umol/L,and 1 umol/L of fucoxanthin,respective-ly.Results:Compared with the blank controls,as the concentration increased,the inhibitory effect of fucoxanthin on HeLa cell growth gradually increased(P<0.05).At the same time,as the dosage and stimulation time of fucoxanthin increased,the apoptosis rate of HeLa cells significantly increased(P<0.05).As the concentration increased,the ATP production rate of HeLa cells stimulated by fu-coxanthin also significantly decreased(P<0.05).Conclusion:Fucoxanthin can inhibit the growth and induce apoptosis of cervical cancer HeLa cells.
