Optimization of Determination of Protein in Activated Sludge by Bradford Method
The protein content in activated sludge was detected by Bradford Method which was optimized through the procedure that from washing of sludge,extraction of protein to detection of protein content.The results showed great linear relationship in range of 10~180 μg/mL of protein (R2=0.995 7) with the operation conditions of washing by Tris-HCl buffer solution,extraction by alkaline,water-bath for 5 ~10 min and detection under neutral,weak-acid or weak-alkaline conditions.The optimized method showed high precision with RSD 3.80%,good reappearance with RSD 2.09% and the average recovery of the additional sample (n=6) was 96.99%~109.14%.This method which is rapid,accurate and reliable,could be applied for the detection of protein content in activated sludge.
