Effect of miR-628-3p targeting ERRa on malignant biological behavior of osteosarcoma cell line MG-63 induced by bisphenol A exposure
Objective To understand the effect of bisphenol A on the proliferation,apoptosis and invasion of osteosarcoma cell line MG-63 based on the miR-628-3p-ERRa pathway.Methods The osteosarcoma cell line MG-63 model was established in vitro.After being treated with bisphenol A at the doses of 0(control),50,100,and 200 μmol/L respectively for 72 h,MTT method,crystal violet staining,flow cytometry,Transwell chamber,and the scratch experiment was used to measure cell viability,the number of monoclonal formation,apoptosis level,cell invasion,and migration horizontal distance.RT-PCR method and Western blot method were used to measure the gene and protein expression levels of cellular miR-628-3p and ERRa.Results The survival rate and number of monoclonal formation of MG-63 cells in the groups exposed to bisphenol A at different concentrations were higher than those in the control group,with significant differences(P<0.01),and as the concentration of bisphenol A increased,the survival rate and the number of monoclonal formation of MG-63 cells showed an upward trend.The invasion number and migration distance of MG-63 cells in the groups exposed to bisphenol A at different concentrations were significantly higher than those in the control group(P<0.01),and with the increase of bisphenol A dose,the invasion number and migration distance of MG-63 cells showed an upward trend.The apoptosis rates of MG-63 cells in different bisphenol A exposure groups were significantly lower than those in the control group(P<0.01),and as bisphenol A exposure increased,MG-63 cells apoptosis rate showed a downward trend.The expression levels of miR-628-3p mRNA in MG-63 cells in different bisphenol A exposure groups were lower than those in the control group,and the expression levels of ERRa mRNA and protein were both significantly higher than those in the control group(P<0.01),both with a dose dependent trend.Conclusion Bisphenol A exposure can inhibit the expression of miR-628-3p in osteosarcoma cancer cells and promote the high expression of ERRa,thereby promoting the proliferation,invasion and migration of osteosarcoma cancer cells and inhibiting their apoptosis.
