化妆品中阿莫罗芬等5种抗感染药物的液相色谱-串联质谱同时测定法

目的 建立液相色谱-串联质谱(HPLC-MS-MS)同时测定化妆品中阿莫罗芬、布替萘芬、舍他康唑、酞丁安、克罗米通等5种抗感染药物的分析方法.方法 样品采用乙腈超声提取,聚四氟乙烯滤膜过滤,再通过LunaC8色谱柱(100 mm×2 mm,3 μm)分离,柱温为30℃,流速为0.3 ml/min,流动相:0.5%甲酸乙腈-0.5%甲酸水.采用电喷雾离子源及多反应监测模式(MRM)对目标化合物进行测定.结果 阿莫罗芬的线性范围在0.004~0.2 μg/ml,布替萘芬和克罗米通的线性范围在0.002~0.1 μg/ml,舍他康唑和酞丁安的线性范围在0.02~1 μg/ml,所得回归方程均呈较好的线性关系,r>0.998.该方法检出限为0.02~0.9 mg/kg,在3类不同基质化妆品中平均加标回收率的范围为87.3%~107.9%,RSD为1.6%~13.4%.结论 该方法高效、快捷、准确,适用于多种化妆品基质中阿莫罗芬等5种抗感染药物的同时快速测定.
Simultaneous determination of 5 anti-infective drugs in cosmetics by HPLC-MS-MS
Objective To develop an HPLC-MS-MS analytical method for simultaneous determination of amorofine,butenafine,sertaconazole,fibamzone,crotamiton and other five anti-infective drugs in cosmetics.Methods The sample was extracted with acetonitrile under ultrasonication,and filtered by PTEF filter membrane.The analytes were separated on Luna C8 column(100 mm×2 mm,3 μm),the column temperature was 30 aC and the flow rate was 0.3 ml/min.The mobile phase consisted of 0.5%formic acid acetonitrile-0.5%formic acid water.The compounds were identified under electrospray ionization and multiple reactions monitoring(MRM)mode.Results The linear ranges of amorofine were 0.004-0.2 μg/ml,that of butenafine and crotamiton were 0.002-0.1 μg/ml,those of sertaconazole and fibamzone were 0.02-1 μg/ml.In the ranges,the obtained regression equations all showed good linear correlation(r>0.998).The LODs of five anti-infective drugs were 0.02-0.9 mg/kg.The recoveries of five anti-infective drugs in 3 different dosage forms of cosmetics ranged 87.3%-107.9%,with RSDs of 1.6%-13.4%.Conclusion This method is effective,simple and accurate,and it is applicable to the simultaneous determination of five anti-infective drugs in various cosmetics substrates in cosmetics.
