Increase of PGC-1α Expression in Mice Liver by Adding Ligustrazine Hydrochloride under Cold Exposure
To investigate the effect of ligustrazine hydrochloride on liver heat producing protein PGC-1α in mice under cold exposure,40 C57BL/6 male mice were randomly divided into four groups after pre-feeding for one week.The normal temperature control group was kept at 26±2℃,and the cold exposure group and the drug addition group were placed in the artificial climate chamber at 4℃ for cold stimulation for 3 h every day.The mice in the drug addition group were given corresponding gavage before cold exposure,and the mice were killed four weeks later and liver tissues were collected.HE staining was used to observe the damage of liver tissue.Oil red O staining was used to observe the number of lipid droplets in liver tissue.The expression level of PGC-1α in liver was determined by IHC and Western Blot.The results showed that compared with the control group,the liver was not damaged after ligustrazine hydrochloride was added,the number of lipid drops was increased,and the expression level of PGC-1α was significantly increased.In conclusion,ligustrazine hydrochloride could increase the expression of PGC-1α and promote liver heat production in mice under cold exposure.
