固定化酶法制备甘油磷酸胆碱工艺的研究

固定化脂肪酶具有安全绿色、特异性强、易于分离的优点,为制备一种高纯度食品级的认知增强剂L-α-甘油磷酰胆碱(Glycerol phosphorylcholine,以下简称GPC),以GPC相对含量为评价指标,探讨了在有机相中酶法水解大豆磷脂制备GPC,考察了反应时间、底物浓度、加水量、加酶量对磷脂水解程度的影响.在单因素试验的基础上,经响应面试验得到制备GPC的最佳工艺条件为:反应时间37.7 h,底物浓度16%,加水量 63%,加酶量 17%,该条件下GPC的相对含量为 78%.进一步经硅胶柱层析纯化,GPC的相对含量可达到 99.8%.试验表明,该方法可实现高纯GPC的有效制备.
Preparation of Glycerol Phosphatidylcholine by Immobilized Enzyme
Immobilized lipase has the advantages of safe green,strong specificity and easy separation.In order to study the preparation of glycerol phosphorylcholine(Glycerol phosphorylcholine,GPC)by enzymatic hydrolysis of soybean phospholipids in organic phase,taking the relative content of GPC as the evaluation index,the effects of reaction time,substrate concentration,water addition and enzyme addition on the degree of phospholipid hydrolysis were investigated.On the basis of single factor test and response surface test,the optimum process conditions for preparing GPC were as follows:reaction time 37.7 h,substrate concentration 16%,water addition 63%,enzyme addition 17%.Under these conditions,the relative content of GPC was 78%.After further purification by silica gel column chromatography,the relative content of GPC could reach 99.8%.Experiments showed that this method could effectively prepare high purity GPC.
