响应面法优化英国红芸豆多糖提取工艺及抗氧化活性研究

Optimization of Extraction Process and Antioxidant Activity of Polysaccharide from British Red Kidney Bean by Response Surface Methodology

以英国红芸豆为原料,多糖得率为考察指标,用响应面试验优化提取工艺,并对英国红芸豆多糖体外抗氧化活性进行分析.结果表明,英国红芸豆多糖最优提取工艺为:提取温度 90℃、提取时间 1.5 h、料液比 1∶40 g·mL-1,在此条件多糖得率最优为4.86%.抗氧化活性试验表明:英国红芸豆多糖的抗氧化活性与其浓度呈量效关系,当多糖浓度为5 mg·mL-1 时,对DPPH自由基、·OH自由基和O2-自由基清除率分别为 69.85%、53.61%和 21.36%.
Taking British red kidney bean as raw material,the extraction process was optimized by response surface experiment,and the antioxidant activity of British red kidney bean polysaccharide was analyzed in vitro.The results showed that the optimal extraction process of polysaccharide from British red kidney bean was as follows:extraction temperature 90℃,extraction time 1.5 h,solid-liquid ratio 1∶40 g·mL-1,and the optimal extraction rate of polysaccharide was 4.86%.The antioxidant activity test showed that the antioxidant activity of British red kidney bean polysaccharide was dose-effect dependent on its concentration.When the concentration of polysaccharide was 5 mg·mL-1,the scavenging rates of DPPH free radical,·OH free radical and O2-free radical were 69.85%,53.61%and 21.36%,respectively.
