Screening of Molecular Markers Associated with ty-5 Gene of Tomato Based on DNA-BSA Resequencing and SSR Technology
The location of disease-resistant gene ty-5 of tomato yellow leaf curl virus was studied by DNA-BSA resequencing and SSR markers.Seven candidate regions related to tomato ty-5 gene were obtained by DNA-BSA resequencing,respectively located on chromosomes 1,4,and 9,with a total length of 21.68 Mb.Two markers,C_2at4g34700 and SSR383,were obtained by SSR markers,which were closely linked to ty-5 gene of tomato leaf leaf curve virus disease resistance,and no single recombinant strain was found.Finally,the results of DNA-BSA resequencing and SSR marker were analyzed jointly,and the markers C_2at4g34700 and SSR383 were located near the disease resistance associated regions of chromosomes 1 and 9 in the resequencing association analysis.It was preliminarily concluded that these two markers could be used as molecular markers of ty-5 gene of tomato against yellow leaf curl virus disease.
