Transcriptome Analysis of BLV-△miRNAs and BLV Infected Bovine Mammary Epithelial Cells
Enzootic Bovine Leukosis(EBL)is a tumor proliferative disease caused by Bovine leukemia virus(BLV)infection,which is widely prevalent in dairy cows in China and restricts the development of dairy farming industry in China.BLV is generally infected through intrauterine infection and ingestion of infected milk and colostrum.The infected bovine mammary epithelial cells are more susceptible to Staphylococcus aureus.As a functional element necessary for efficient replication of BLV,BLV-miRNAs play a key role in the process of tumor formation,but the effect of BLV-miRNAs on bovine mammary tissue is not reported.Bovine mammary epithelial cells were infected with BLV and BLV-△miRNAs,respectively,and UMI absolute quantitative transcriptome sequencing was performed.The results showed that BLV and BLV-△miRNAs infected bovine mammary epithelial cells changed the level of gene transcription.The differentially expressed genes were mainly enriched in biological processes,including gene transcription(HMGA1,C1QBP,PTMA,etc.),cell senescence(HMGA1,SERP1,etc.),cell proliferation and migration(PTPRJ,EPPK1,etc.),viral replication(HMGA1,C1QBP,etc.),protein metabolism(SERP1,USE1,etc.),immune function(PTMA,PTPRJ,etc.).KEGG analysis showed that BLV-miRNAs could activate metabolic pathways,miRNA and cancer,viral infection,viral carcinogenesis,IL-17 signaling pathway,adhesion connection and other signaling pathways.Therefore,BLV-miRNAs might affect the immune function of mammary epithelial cells and change the cell state when BLV infected the mammary gland tissue of dairy cows,thus accelerating the process of viral in-fection,promoting the infection of pathogens and accelerating the occurrence and development of mammary gland diseases in dairy cows.The results laid a foundation for further study on the function of BLV-miRNAs.
