Drug Efflux of E.coli ABC Superfamily Transporter YadGH and Verification of Transported Drug by YadGH
ABC transporters mediate drug resistance in Escherichia coli.However,the drug resistance mediated by E.coli ABC transporter YadGH and the type of drug transported has not been reported.To explore the efflux function of YadGH transporter,the bioinformatics analysis of YadGH was carried out.Then yadG gene knockout strains of E.coli K-12 were constructed by Red recombinant technology,and the minimum inhibitory concentration of antibiotics against the yadG gene knockout strains and the ATPase activity of the purified YadG protein were measured.The results showed that the efflux ability of the knockout strains to transport roxithromycin,tylosin,and erythromycin was decreased,and YadG protein showed ATPase activity.The results demonstrated that YadGH was an ABC transporter of macrolide.
