摘要

进入21世纪后,微生物DNA提取方法发展到了新高度,但人们对部分微生物的研究仍处于摸索阶段,如人类痤疮、黑鼻头等皮肤微生物.为了进一步优化其DNA提取方法,以痤疮毛囊微生物为样本,采用两种方法(CTAB法和DNeasy Blood & Tissue试剂盒)进行DNA提取,对DNA的纯度、得率、PCR扩增及凝胶电泳等进行比较研究.结果表明,两批样本中夏季批CTAB法的DNA平均得率分别为1024.83和1.40,冬季批CTAB法的DNA平均得率分别为383.40和1.09,两种方法均可用于痤疮毛囊微生物的DNA提取.但从PCR扩增后分析来看,试剂盒法具有提取效果较好、针对性强、耗时较短及方便的优点,是从痤疮毛囊中提取微生物DNA的良好方法.

Abstract

In the 21st century,the methods of extracting microbial DNA have developed to a new level,but the research on some microorganisms is still in the exploratory stage,such as human acne,black nose and other skin microorganisms.In order to further optimize their DNA extraction methods,two DNA extraction methods(CTAB method and DNeasy Blood & Tissue)are used to extract DNA from acne follicle microorganisms.The purity and yield of DNA,PCR amplification and gel electrophoresis are compared.The results show that the average DNA yield of CTAB method in summer and winter are 1 024.83 and 1.40,respectively.The average DNA yield of CTAB method in winter are 383.40 and 1.09,respectively.Both two methods can be used to extract DNA of acne hair follicle microorganisms.From the analysis after PCR amplification,the kit method has the advantages of good extraction effect,strong pertinence,short time and convenience,so it is the good method of DNA extraction of method of acne hair follicle microorganisms.
