The establishment of a rat model of chronic visceral hypersensitivity(VHL)and intestinal high expression of TRPV1 can lay a foundation for the research and development of targeted therapeutic drugs for patients with IBS.The study constructs model rats with Capsaicin(CAP)as stimulant,evaluates the intestinal sensitivity and TRPV1 expression of rats with different concentrations of CAP through abdominal regression reflex(AWR)score,RT-PCR and western blot.The results show that after continuous administration of 0.1%~0.5%CAP,the AWR score of the experimental group is higher than that of the blank group.There is no significant difference in AWR scores between the interval stimulation group and the continuous stimulation group.But the results of HE staining show that there is no significant difference in AWR scores between the interval stimulation group and the continuous stimulation group.RT-PCR and western blot analysis show that the jejunum,ileum and colon of the experimental group of rats injected with 0.1%~0.3%CAP have enhanced TRPV1 expression to a certain extent.Through the condition analysis of different concentrations of CAP and stimulation intervals,and the combination with the results of intestinal tissue HE staining and TRPV1 expression,the study finally selects 0.3%CAP to intervel stimulation for 1 day and last 14 days as the best model of chronic VHL and high expression of TRPV1 in rats.
