Comparison of the Embellishment of Subcutaneous Tumor Model of Hepatocellular Carcinoma with Two Methods/
Objective:To establish subcutaneous tumor mass model of Wistar and BALB/c nude mice by injecting Walker-256 tumor cells.Methods:The ascites of BALB/c nude mice containing Walker-256 tumor cells were extracted,centrifugally washed,and the washing solution was inoculated into the subcutaneous inguinal region of Wistar mice and BALB/c nude mice to form subcutaneous tumor masses.The tumor formation rate and tumor growth rate of the subcutaneous tumor mass model were observed and compared between two different experimental mice.After experiment,the microvascular density(MVD)and the expression of vascular endothelial growth factor(VEGF)were detected by immunohistochemistry.Results:Success rate of both methods were 100%.The growth rate of tumor mass in BALB/c nude mice was faster than that in Wistar rats.And the molecular expression of VEGF and MVD in the group of BALB/c nude mice were both higher than those of Wistar rats(t=8.52,5.13;P<0.05).Conclusion:Both Wistar mice and nude mice can meet the needs of clinical trials and studies.However,as nude mice are immunodeficient animals,the growth rate of tumor mass is significantly faster than that of Wistar mice,and the tumor angiogenesis index is higher.Therefore,nude mice are the best choice when choosing the growth model of liver cancer tumor mass.
