Objective:In order to obtain the optimal extraction process conditions and quality detection method for polysaccharides from Lepista nuda.To investigate the optimum extraction process of polysaccharides from Lepista Sordida Sing and to analyse the quality of polysaccharides by using a variety of chromatographic methods.Methods:Orthogonal method to optimize extraction process,Column chromatography purification,UV、HPLC、IR、HPCE Spectra Analysis.Results:Opimal process of extraction procedure of poly-saccharides from Lepista Sordida Sing were obtained:temperature 100℃、extraction time 10h、material liquid ratio 1:25;two components Ⅰ and Ⅱ was separated by Column chromatography,Ⅰ is the main component.UV results show that the polysaccharide from Lepista Sordida Sing does not contain nucleic acids and proteins.HPLC results showed that there was a significant absorption peak of polysaccharide in Ⅰ component.IR results show that several typical characteristic absorption peaks of polysaccharide within 400~4000cm-1 which proved the polysaccharides is complex polysaccharides connected by a variety of glycosidic bond of sugar.HPCE results show that the polysaccharides consists of six simple sugars including glucosamine and glucuronid acid.Conclusion:The structure and properties of polysaccharides are studied by UV,HPLC,IR,HPCE chromatographic methods,which has important guiding significance for the development of new polysaccharide health foods and medicines.
