基于VEGF-VEGFR信号通路探讨补肺消结汤对肺癌大鼠的机制研究

目的:探讨补肺消结汤基于血管内皮生长因子(Vascular endothelial growth factor,VEGF)或VEGF受体-2(Vascular endothelial growth factor receptor-2,VEGFR-2)信号通路对肺癌大鼠的具体作用机制.方法:本研究通过建立A549肺癌大鼠移植瘤模型90例进行实验,随机分为对照组(n=30)、补肺消结汤低剂量组(n=30)及高剂量组(n=30),补肺消结汤低剂量组、高剂量组分别予以10mL/kg及20mL/kg剂量灌胃,对照组给予等量生理盐水灌胃,每组1次/天,连续持续14天.对肺组织进行病理组织学观察.同时,采用实时荧光定量PCR(Quantitative real-time PCR,qRT-PCR)检测肺癌组织中p53 mRNA表达情况,并用免疫印记方法(Western blot)检测大鼠肺组织中VEGF、VEGFR-2表达.结果:与对照组相比,低、高剂量组p53 mRNA水平表达显著升高(P<0.05).与对照组相比,低、高剂量组VEGF、VEGFR-2表达显著降低(P<0.05).结论:补肺消结汤可通过调控VEGF-VEGFR信号通路的基因表达,提高p53 mRNA表达水平,抑制大鼠的肺癌生长.
Study on the Mechanism of Bufeixiaojie Decoction on Lung Cancer Rats Based on VEGF-VEGFR Signaling Pathway
Objective:To investigate the Vascular endothelial growth factor(VEGF)or VEGF receptor-2(Vascular endothelial growth factor receptor-2)based on Bufei Xiaojie Decoction.The specific mechanism of VEGFR-2 signaling pathway on lung cancer rats.Methods:In this study,30 cases of transplanted tumor model of A549 lung cancer rats were established and randomly divided into control group(n=30),Bufei Xiaojie Decoction low-dose group(n=30)and high-dose group(n=30).Bufei Xiaojie Decoction low-dose group and high-dose group were given gavage of 10mL/kg and 20mL/kg,respectively.The control group was given the same amount of normal saline intragastrically,once a day in each group,for 14 days.The lung tissue was observed by histopathology.Meanwhile,Quantitative real-time PCR(qRT-PCR)was used to detect p53 mRNA expression in lung cancer tissue,and Western blot was used to detect VEGF and VEGFR-2 expression in rat lung tissue.Results:Compared with control group,p53 mRNA level in low and high dose groups was significantly increased(P<0.05).Compared with the control group,the expressions of VEGF and VEGFR-2 in low and high dose groups were significantly decreased(P<0.05).Conclusion:Bufei Xiaojie Decoction can regulate the gene ex-pression of VEGF-VEGFR signaling pathway,increase the expression level of p53 mRNA,and inhibit the growth of lung cancer in rats.
