目的:探讨POH1在甲状腺乳头状癌(PTC)中的表达、临床意义及其在癌细胞增殖中的作用.方法:本研究通过免疫组化、qRT-PCR和Western blot等方法分析POH1在PTC中的表达,并建立POH1的表达与PTC患者的临床病理参数联系.采用MTT、CCK8等技术,通过SiRNA敲降POH1检测其对PTC细胞增殖的影响.结果:POH1表达和PTC患者性别、年龄、瘤体数目等均无关(P>0.05),而与T分期相关(P<0.05).POH1在PTC组织中表现出高表达,而在癌旁组织中低表达甚至不表达.转染24h、48h后,Si-POH1细胞增殖活性显著低于Si-NC.结论:POH1能促进PTC细胞增殖,有望成为PTC临床诊疗及预后评估的分子标志物.
Objective:To investigate the expression and clinical significance of POH1 in papillary thyroid carcinoma(PTC)and its role in the proliferation of cancer cells.Methods:In this study,immunohistochemistry,qRT-PCR and Western blot were used to detect the expression of POH1 in PTC and paracancer tissues,and the relationship between POH1 expression and clinicopathological parameters of PTC patients was analyzed.MTT and CCK8 techniques were used to detect the effect of SiRNA knockdown POH1 on the proliferation of PTC cells.Results:POH1 expression was not correlated with sex,age,tumor number and lymph node metastasis of PTC patients(P>0.05),but was correlated with T stage(P<0.05).POH1 is highly expressed in PTC tissues,but low or even no expression in paracancer tissues.The proliferative activity of Si-POH1 cells was significantly lower than that of Si-NC cells after transfection for 24h and 48h.Conclusion:POH1 can promote the proliferation of PTC cells and is expected to be a molecular marker for clinical diagnosis,treatment and prognosis evaluation of PTC.
