Identification of Saprophagous Aphid-Eating Flies Based on Molecular Biology
This study aimed to identify four species of saprophagous aphid-eating flies collected in Changsha.The morphological characteristics of the flies were observed,and the genomic DNA was extracted.The mitochondrial cytochrome C oxidase subunit I(COⅠ)gene fragments were amplified by PCR and sequenced.The sequencing results were compared with the sequences in GenBank,and a phylogenetic tree was constructed with the neighbor-joining method.The results showed that the four species of flies presented consistent morphological characteristics with the target flies.The COⅠ genes of the four species showed high homology with those in GenBank.The phylogenetic tree constructed based on COⅠsequences showed that Eristalis tenax and Eristalis cerealis shared the same clade.Considering both morphological characteristics and molecular biology,the four species of saprophagous aphid-eating flies captured in Changsha were identified as Phytomia zonata,Eristalis cerealis,Eristalis tenax,and Eristalinus arvorum,which laid a foundation for subsequent research.
