Cloning and Sequence Analysis of the Gene of MYB Transcriptional Regulator from Lolium perenne L.
To further study the function of the MYB in stress resistance and development in Lolium perenne L. , the conserved region of MYB transcription factor which is related with resilience and development was amplified by PCR using homologous method and cloned from leaves of the Lolium perenne L. Sequence analysis and function prediction were performed as well. Sequencing results showed that three different gene fragments of MYB transcription factor were cloned and named separately as LpMYBl ,LpMYB2 and LpMYB3.The DNA lengths of LpMYBl ,LpMYB2 and LpMYB3 were 221 bp,221 bp and 202 bp,which included 103 bp,103 bp and 84 bp introns, respectively. All of them encoded 39 amino acids. Sequence homologous comparison showed that these three genes might be involved in adversity response and morphogenesis of Lolium perenne L. Phylogenetic analysis indicated that phylogenetic development of the three genes was consistent with the evolutionary model of monocotyledons. The results showed that the three MYB genes were involved in the expression of resistance and development of Lolium perenne L. , which provides a foundation for study of function of MYB transcription factor gene in Lolium perenne L.
