Construction of Fingerprinting and Analysis of Genetic Diversity with ISSR Markers for 22 Sweet Potato Varieties
The DNA fingerprinting of 22 sweet potato varieties were constructed using ISSR makers,and genetic diversity was analyzed.From 40 pairs of ISSR primers, 12 pairs of primers were selected to amplify, and 94 bands were obtained ,on the average ,each pair of primer was amplified to 7.83 bands ,69 polymorphic bands were obtained,the polymorphic percentage of bands was 73.40%.By combining three pairs of primer UBC8, UBC35 (1) and UBC41,22 sweet potato species fingerprinting were constructed.The cluster analysis showed that 22 sweet potato species could be classified into four kinds when threshold was 23, among them ISSR analysis of partial species agreed with pedigree, or related to geography places.
