Identification and Biological Characterization of the Pathogen of Allium ascalonicum Gray Mould
In order to identify the pathogen and its biological characteristics of Allium ascalonicum gray mould,the pathogen was isolated from diseased leaves of A.ascalonicum by single spore isolation method.Based on the morphological characteristics and rDNA-ITS sequence analysis,the pathogen of A.ascalonicum gray mould was identified as Botrytis cinerea Pers..The biological characteristics of B.cinerea were studied,and the results showed that the pathogen was able to grow on the ten media tested,during which the optimum medium for the growth of the pathogen was PDA medium.In the nine carbon sources and eleven nitrogen sources tested,the optimum carbon and nitrogen sources were soluble starch and yeast powder respectively.The pathogen could grow in pH from 4.0 to 12.0,while the optimal pH was 5.0-6.0.The pathogen could grow at 4-30 ℃,and 15-25 ℃ was suitable for the mycelia growth.The mycelia grew quickly and very densely at 25 ℃,when the temperature was above 30 ℃,the mycelia growth stopped.The mycelia lethal temperature was 52 ℃ for 10 min.
